A quick look at my local mail box and its permissions are 770.
I assume you have all the directories required by maildir?
/home/mailer/Maildir/tmp/
/home/mailer/Maildir/cur/
/home/mailer/Maildir/new/
Some default setups point to .Maildir instead of Maildir
